What Is Acetocarmine Stain Used For?
The onion root tip squash is the classic practical. Freshly grown root tips are fixed, softened in warm dilute hydrochloric acid so the cells separate, and then warmed gently in a few drops of acetocarmine in a watch glass over a spirit lamp without boiling. The tip is moved to a slide, covered with a coverslip and pressed firmly to spread the cells into a single layer. Under the microscope the chromosomes stand out clearly, and students can identify prophase, metaphase, anaphase and telophase in the same field.
Young anthers from flower buds are squashed the same way to show stages of meiosis, and college classes also use the stain for salivary gland chromosomes. Comparing a student's temporary slide with a reference slide of the same tissue is a good way to check technique. The stain contains acid, so use it in a ventilated area, keep it off skin and clothing, and wipe the underside of the slide before it goes on the microscope stage.
