What Is the Protease Enzyme Used For?
In a classroom DNA separation, crushed fruit, onion or cheek cells are mixed with a salt and detergent solution to open the cells. A small amount of protease is then stirred in and the mixture is left to stand, often in a warm water bath, while the enzyme digests the proteins that hold DNA in a tightly packed form. After filtering, chilled alcohol is poured gently down the side of the tube, and the freed DNA gathers at the boundary as a white, stringy mass that students can lift out.
Protease also makes a clear stand-alone demonstration of enzyme action. Added to a cloudy egg-white suspension or to milk, it gradually changes the protein, and students can confirm the breakdown by showing a weaker biuret result or a stronger ninhydrin result as digestion proceeds. Setting tubes at different temperatures and including a boiled-enzyme control turns the activity into a complete investigation, similar to those in an enzyme activity kit for biology classes.
