What Is the Oxidase Reagent Used For?

The oxidase test asks one quick question of a bacterial colony: does it produce cytochrome c oxidase? A little of the colony is rubbed on to filter paper moistened with the reagent, or the reagent is dropped on to colonies, and a colour change signals an oxidase-positive organism. It is one of the first tests used to sort Gram-negative rods, and it also features in water testing microbiology, where presumptive coliforms are expected to be oxidase-negative.

Pick colonies with a platinum loop, a plastic loop or a wooden stick, because nichrome or iron wire can give a false-positive reaction. Test fresh growth, read the result within the time stated in the instructions supplied with the reagent, and run known positive and negative cultures alongside. Many oxidase reagents darken on exposure to air and light, so keep the bottle tightly closed and cool between sessions.
