Transpiration is not the same as simple evaporation. Water moves from the soil into the roots, up the stem through the conducting vessels and out through the leaves into the air, and the rate depends both on the surroundings, such as light, temperature and air movement, and on the plant itself, which regulates the loss through its stomata. In young plants almost every surface exposed to air can lose water this way, which is why transpiration is treated as a complex physiological process rather than a purely physical one.

In class, a leafy shoot is placed in the coloured liquid, held by the stand and clamp, and students watch the colour move into the veins while the liquid level falls. Comparing a shoot in still air with one near a fan, or one with its leaves coated in petroleum jelly, turns the demonstration into a fair-test investigation for botany lab equipment practicals. Cut shoots under water to avoid air locks, and rinse the glass tube and beaker straight after use so the colouring does not stain them.
