In the test for starch in a leaf, the leaf is first softened in boiling water, then placed in a tube of ethanol standing in the hot water bath until the green pigment dissolves out; after rinsing, iodine solution shows which parts of the leaf made starch. In DNA isolation, ice-cold ethanol poured gently onto the extract makes DNA come out of solution as white threads. For permanent slides, ethanol diluted with distilled water into a graded series gradually removes water from stained sections, pairing naturally with the stains of a staining kit for the biology lab in India.
Ethanol is highly flammable: never heat it directly over a flame, switch off burners before opening the bottle, and warm it only in a water bath heated beforehand.
