Eosin is best known as the second half of haematoxylin and eosin (H and E) staining, the combination used to study tissue sections in histology courses. After sections are stained with haematoxylin and blued, the slides pass briefly through eosin, are rinsed, and are taken through graded alcohols before clearing and mounting. The result shows purple-blue nuclei against pink cytoplasm and fibres, the view most textbooks use for epithelium, muscle, liver or kidney sections.
Students preparing permanent slides soon learn that timing matters: a longer dip or less time in the alcohols gives a deeper pink, while extra rinsing lightens it. Eosin can also be used on its own for a quick temporary mount of cheek cells or to highlight cytoplasm in simple animal tissue preparations. Use a staining jar so several slides are treated evenly, keep the jar covered between runs, and wipe drips promptly, because the dye marks benches and fingers.
