The sample is first diluted in a suitable diluting fluid, a flat cover glass is placed across the raised supports, and the chamber is filled from the edge by capillary action. Once the cells settle, the technician counts them in defined squares of the grid under the microscope. Because the ruled area and the 0.1mm depth fix the volume above each square, the count can be converted into cells per unit volume by allowing for the squares counted and the dilution used. The double grid lets two separate fillings, or duplicate counts of one sample, be read on the same chamber.
